| We are using this phosphoproteomics approach, to look at signaling events downstream of insulin binding to its receptor, both in vitro and in vivo. Preliminary results from Chinese hamster ovary (CHO) cells over expressing the insulin receptor show us some of the proteins we expect to be tyrosine phosphorylated in response to insulin, like insulin noble feeder receptor and IRS1. These cells noble feeder are being further interrogated to find novel tyrosine phosphorylation events that mediate insulin signaling. These studies are being extended to the whole animal noble feeder by taking livers for insulin-treated animals and subjecting them to the same phosphoproteomics analysis. To begin to characterize the insulin resistant state, we are profiling the mouse's response to insulin at the mRNA level, both in normal animals and in animals genetically modified to be insulin resistant. |
